Description:

Mannitol salt agar (MSA) with streptomycin sulfate is a modified form of standard MSA in which an aminoglycoside antibiotic is added to increase the suppression of streptomycin-susceptible background bacteria, while retaining the usual selectivity for salt-tolerant staphylococci and the mannitol/phenol-red differentiation system.  MSA contains peptones and beef extract, which provide nitrogen, amino acids, vitamins, minerals and other growth factors needed for bacterial growth, NaCl creates high-salt conditions and most bacteria are unable to grow under these conditions.  D-mannitol is a fermentable carbohydrate in the medium, and phenol red is the pH indicator with the medium appearing red at neutral to alkaline pH and turns yellow when acid is produced from the mannitol fermentation.

Streptomycin is an aminoglycoside antibiotic that binds to the 30S ribosomal subunit in susceptible bacteria, causing misreading of mRNA, inhibition of protein synthesis, and eventual cell death.  Streptomycin is active against aerobic Gram-negative rods and some Gram-positive bacteria.  The combination of MSA and Streptomycin sulfate introduces an additional antibiotic selection layer, preferentially inhibiting streptomycin-susceptible organisms, that might otherwise still grow under high-salt conditions.
