Description:
Mannitol Salt Agar (MSA) with penicillin G is a modified version of standard MSA in which a β-lactam antibiotic is added to further suppress susceptible background bacteria, while preserving the usual selectivity for salt-tolerant bacteria and the mannitol-based differentiation system.
MSA provides peptones and beef extract that supply nitrogen, amino acids, vitamins minerals necessary for bacterial growth. NaCl creates high salt conditions that most bacteria are unable to tolerate, enriching salt-tolerant organisms such as staphylococci. D-mannitol is a fermentable sugar, and phenol red acts as a pH indicator, that is red at neutral/alkaline pH and yellow when acid is produced from mannitol fermentation. The addition of Penicillin G which is a β-lactam antibiotic targets cell-wall synthesis in susceptible bacteria by binding penicillin-binding proteins and blocking peptidoglycan cross-linking, which leads to the weakening of the cell wall, osmotic instability and lysis.
