Description:

Luria-Bertani (LB) agar with tetracycline is a rich bacterial growth medium (standard LB agar) that has been made selective with the addition of the antibiotic tetracycline.  This allows the growth of tetracycline-resistant bacteria while inhibiting susceptible ones.  LB base provides peptides, amino acids, vitamins, and salts that support the growth of non-fastidious bacteria such as E.coli.

Tetracycline is a broad-spectrum antibiotic that binds to the 30S ribosomal subunit and blocks the attachment of aminoacyl-tRNA to the ribosomal A site.  This causes protein syntheis to be inhibited.  Bacteria that are tetracycline resistant can grow and form colonies on the LB tetracycline agar, whereas susceptible cells show strongly inhibited growth.  LB agar with tetracycline is often used as a selective medium for maintaining plasmids that carry tetracycline-resistance markers in E. coli and other lab strains, selecting transformants or recombinants that have acquired tetracycline-marked plasmids or gene cassettes, and enriching or isolating tetracycline-resistant bacteria while suppressing tetracycline-susceptible flora.
