Description:
Luria–Bertani (LB) agar with Penicillin G is a rich bacterial growth medium (standard LB agar) supplemented with a β-lactam antibiotic to inhibit susceptible bacteria; it is less common than LB with ampicillin for molecular cloning but is conceptually similar in how it works.
The LB base provides peptides, amino acids, vitamins, and salts that support robust growth of non-fastidious bacteria such as E. coli. Penicillin G is a β-lactam antibiotic that targets cell wall synthesis (binding penicillin-binding proteins and blocking peptidoglycan cross-linking), leading to lysis and death of actively growing, susceptible bacteria, especially many Gram-positives. Bacteria with appropriate β-lactam resistance mechanisms (e.g., β-lactamase production or altered PBPs) can grow and form colonies on LB-Pen G; susceptible cells are inhibited or killed. LB agar with Penicillin G can therefore be used as a selective medium to suppress penicillin-susceptible bacteria (commonly Gram-positives) when enriching for penicillin-resistant bacteria or focusing on Gram-negative flora.
