Description:
Luria–Bertani (LB) agar with gentamicin is a rich, non-selective bacterial growth medium (LB agar) that has been made selective by adding the aminoglycoside antibiotic gentamicin. It is used to grow and maintain bacteria that carry gentamicin-resistance genes (e.g., plasmid-encoded gentamicin resistance), while inhibiting non-resistant cells.
Gentamicin binds the 30S ribosomal subunit and inhibits protein synthesis; in susceptible bacteria, this leads to bactericidal activity. Bacteria that carry a gentamicin-resistance determinant (often a plasmid?encoded aminoglycoside?modifying enzyme) can grow and form colonies on LB agar with gentamicin. LB with gentamicin is a selective medium used for maintaining plasmids with gentamicin resistant markers, selecting transformants or transconjugants containing gentamicin-resistance cassettes, and routine cloning and genetic manipulation where gentamicin is the selectable marker.
