Description:

Luria-Bertani (LB) agar with chloramphenicol is a rich bacterial growth medium (standard LB agar) that has been made selective with the addition of Chloramphenicol typically used to grow and maintain chloramphenicol-resistant E.coli strians and other resistant bacteria the combination of the nutrient richness of LB with the inhibitory activity of chloramphenicol against susceptible cells.

LB supplies peptides, amino acids, vitamins, and salts that support robust growth of non-fastidious bacteria such as E.coli and other related strains.  NaCl maintains osmotic balance and provides essential ions for membrane transport.  Chloramphenicol is a broad-spectrum, primarily bacteriostatic that binds the 50S ribosomal subunit and inhibits peptidyl transferase activity, thereby blocking peptide bond formation and protein synthesis in susceptible bacteria.  Bacteria that carry chloramphenicol resistant-determinants grow and form colonies on LB-chloramphenicol agar; susceptible cells are inhibited or killed.  The combination of LB agar and chloramphenicol as a selective medium allows the cultivation and maintenance of chloramphenicol-resistant E.coli strains used in cloning, expression, or library maintenance.  Maintaining plasmids that carry chloramphenicol-resistance markers and selecting transformants or recombinants containing chloramphenicol-marked plasmids or cassettes.  Selective recovery or enrichment of chloramphenicol-resistant bacteria from mixed samples while suppressing chloramphenicol-susceptible flora.
