Description:
Luria-Bertani (LB) agar with carbenicillin is a rich bacterial growth medium (standard LB agar) that has been made selective from the addition of β-lactam antibiotic carbenicillin, most commonly to grow and maintain plasmid-bearing, carbenicillin-resistant E.coli used in cloning and protein expression. The LB base provides peptides, amino acids, vitamins, and salts that support robust growth of non-fastidious bacteria such as the previously mentioned E.coli.
Carbenicillin is a β-lactam antibiotic that inhibits cell wall synthesis and can be bactericidal at effective concentrations. Ampicillin and carbenicillin are in a similar spectrum, with carbenicillin being more stable, which can lead to fewer satellite colonies being found. LB Agar with carbenicllin can be often seen being used for maintaining plasmids that encode β-lactam resistance in E. coli, and selecting transformants or recombinant clones carrying carbenicllin resistant plasmids in routine cloning and protein-expression workflows.
