Description:
Luria Bertani (LB) agar with ampicillin sodium salt is a rich bacterial growth medium rendered selective by adding ampicillin, used to grow and maintain baceria carrying an ampicilln-resistance gene (usually β-lactamase on a plasmid).
The LB base provides peptides, amino acids, vitamins and salts that support robust growth of non-fastidious bacteria. Ampicillin being a β-lactam antibiotic, inhibits cell wall synthesis and is a bactericidal for susceptible bacteria. Bacteria expressing a β-lactamase can grow and form colonies on LB-amp agar; non-resistant cells are inhibited or die. LB agar with ampicillin therefore is a selective medium widely used for maintaining plasmids with ampicillin resistant markers, selecting transformants or transconjugants carrying ampicillin-resistant cassettes, and routine cloning and genetic manipulation where ampicillin is the selectable marker.
