Dulbecco's Modified Eagle Medium (DMEM) is one of the most widely used modifications of Eagle's medium. DMEM is a modification of Basal Medium Eagle (BME) that contains four fold concentrations of amino acids and vitamins. Additionally, the formulation also includes glycine, serine and ferric nitrate. The original formulation contains 1 g/L of glucose and was originally used to culture embryonic mouse cells. The low glucose level is suitable for growing cells that may be adversely affected by high glucose concentrations, such as certain primary cells, stem cells, or transformed cell lines.
No glucose is to provide a basal medium for studying the metabolic activity and response of cells under low or no glucose conditions. This formulation allows researchers to investigate how cells adapt and function in environments with limited glucose availability, which is particularly relevant for research areas like cancer and stem cell biology.
HEPES, a zwitterionic buffer having a pKa of 7.3 at 37°C prevents the initial rise in pH that tends to occur at the initiation of a culture and increases the buffering capacity to the medium. High glucose may induce ROS, protein modifications, glycosylations and other signaling in certain cell types and in such case, low glucose DMEM may be preferred media. As for glucose contents, DMEM High Glucose has 4.5g/L; MEM has 1g/L; RPMI has 4g/L. Different glucose concentration may affect cell signaling pathways.
This product is 1X Dulbecco's Modified Eagle Medium L-glutamine and 25mM HEPES. It does not contain glucose, sodium pyruvate, sodium bicarbonate nor phenol red. Users should review the literature for recommendations regarding medium supplementation and physiological growth requirements for different cell lines and conditions.

Quality Control
- Apperance: Pale Yellow-colored, clear solution.
- pH (at 1X concentration): 7.4
- Osmolality in mOsm/Kg H2O (at 1X concentration): 310.00 - 350.00
- Sterility: Filtered 0.1 micron twice and 0.04 micron once in a sterile environment. No bacterial or fungal growth is observed after 14 days of incubation, as per USP specification.
- Cultural Response: The growth promotion capacity of the medium is assessed qualitatively by analyzing the cells for morphology and quantitatively by estimating the cell counts and comparing it with a control medium.
- Endotoxin Content: NMT 1EU/ml
Storage and Shelf Life
- Storage: 2-8°C, away from bright light.

- Use before the expiry date given on the product label.
* All DiagnoCine Precision Sterile media are filtered-sterilized with a 0.1-micron filtration two times and 0.04 micron once. Thus, mycoplasma contamination is prevented. The smallest size mycoplasma type can be about 0.2 microns.
