FluxMPS™ RPMI 1640 Medium w/o Sodium Pyruvate, Sodium Bicarbonate
FluxMPS™ DCP-RPMI-PB1X is a Microfluidics Suitable, quadruple-stage ultra-filtered RPMI 1640 formulation engineered for hematopoietic cells, lymphocytes, and related immune-cell models on organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration. Formulation: [+] L-Glutamine, [+] Phenol Red, [+] Calcium, [+] Magnesium, [+] Glucose (2.0 g/L) | [-] Sodium Bicarbonate, [-] Sodium Pyruvate.
- Glucose source: 2000 mg/L (2.0 g/L) D-Glucose — the sole defined carbon source in this bicarbonate-free formulation
- L-Glutamine included at 300 mg/L; glutathione (reduced, 1.0 mg/L) provides additional antioxidant/redox support for lymphocyte and hematopoietic cultures
- Sodium bicarbonate and sodium pyruvate both excluded — CO₂-independent base intended for HEPES (15–25 mM) supplementation for pH control
- Quadruple-stage filtration (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final pore size, with mycoplasma-retentive filtration applied at every stage
- Endotoxin release specification: < 0.05 EU/mL (LAL, USP <85>), controlled per manufacturing batch
- Phenol red (5.3 mg/L) retained for visual pH monitoring; appearance is an orange-colored, clear solution
- Manufactured under an ISO 13485:2016 quality management system; final QC at Diagnocine, Totowa, NJ
- pH 7.4 (USP <791>); osmolality 230–270 mOsm/kg H₂O (USP <785>)

Engineered where standard media fails
Conventional 0.22 µm–filtered RPMI passes mycoplasma-sized organisms, subvisible particulates, and endotoxin fragments that activate TLR4 and confound immune-cell assays. FluxMPS™ is built to reduce those risk factors through a validated, multi-stage filtration process.
Microchannel-safe purity
0.04 µm final filtration; USP <788> particulate compliance. Low-particulate media for suspension immune-cell OoC and flow cytometry workflows.
Immune cell–optimized formulation
RPMI 1640 contains glutathione (reduced) and a balanced profile of vitamins and amino acids suited to lymphocyte and hematopoietic cell culture.
Ultrapure-grade water
Type 1 water, 18.2 MΩ·cm, with tightly controlled trace-metal and organic-carbon (TOC) content — minimizing lot-to-lot variability introduced by feed-water quality.
Batch-tested endotoxin control
Release specification < 0.05 EU/mL (LAL, USP <85>), tested per manufacturing batch — reduces the risk of LPS-driven TLR4 activation artefacts in T cell and NK cell assays.
Rich nutrient profile
RPMI's specialized nutrient set for demanding immune cell types, delivered through a low-particulate filtration process for OoC use.
Customization on demand
pH, nutrient concentrations, HEPES, and component modifications available. Contact support@diagnocine.com.
Quadruple-stage filtration system
Four serial filtration stages — two dedicated prefilter/final-filter pairs — reaching a final 0.04 µm polish under ISO Class 5 aseptic conditions.
-
1
0.1 µm Prefiltration I — Large Particulate Removal
Removes large aggregates and cell debris; protects the first 0.04 µm final filter cartridge.
-
2
0.04 µm Final filtration I — Mycoplasma-Retentive Filtration
Retains particulates in the mycoplasma size range (0.2–0.3 µm) that pass a standard 0.22 µm filter.
-
3
0.1 µm Prefiltration II — Second-Pass Protection
A second, dedicated prefilter protecting the second 0.04 µm final filter cartridge.
-
4
0.04 µm Final filtration II — Polish
Ultimate polishing filter ahead of aseptic fill & finish in an ISO Class 5 (Class 100) environment.
