FluxMPS™ RPMI 1640 Medium with 25mM HEPES w/o Sodium Pyruvate: 1X Liquid
FluxMPS™ DCP-RPMIH-P1X is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) RPMI 1640 + 25mM HEPES formulation engineered for hematopoietic and lymphocyte cell models on organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. The train reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration. HEPES (25 mM, pKa 7.3 at 37°C) supplements the sodium bicarbonate buffering system for pH stability during open-bench handling. Sodium pyruvate is withheld by design so researchers can define their own carbon/OXPHOS substrate levels.
- High-glucose (2 g/L) RPMI 1640 base with 25 mM HEPES (pKa 7.3 at 37°C) for robust pH buffering during open-bench handling and flow cytometry prep
- Sodium pyruvate omitted by design — researcher-defined addition (typically 1 mM) supports OXPHOS and metabolic flux studies in T cells and lymphoma lines
- Reduced glutathione (1 mg/L) included for redox support in lymphocyte and hematopoietic cell culture
- Quadruple-stage filtration (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off — five times finer than 0.22 µm conventional filtration
- Endotoxin release specification < 0.05 EU/mL (LAL, USP <85>) to minimize TLR4-mediated activation artifacts in immune cell assays
- Manufactured under an ISO 13485:2016 quality management system with a per-lot Certificate of Analysis
- Phenol red included (5.3 mg/L) for visual pH monitoring; a phenol red-free variant is available on request for autofluorescence-sensitive imaging

Engineered where standard media fails
Conventional 0.22 µm–filtered RPMI passes mycoplasma-sized particles, subvisible particulates, and endotoxin fragments that can confound sensitive immune cell assays. FluxMPS™ is built around a finer filtration train to reduce these risks.
Microchannel-safe purity
0.04 µm final-stage filtration with USP <788> Method 1 particulate compliance (NMT 25/mL ≥10 µm, NMT 3/mL ≥25 µm) — supports suspension immune cell OoC and flow cytometry workflows.
Total metabolic control
Sodium pyruvate is withheld so researchers can define their own carbon and OXPHOS substrate levels for T cell and lymphoma metabolic flux studies, without a pre-set pyruvate background.
Ultrapure-grade water
Prepared with Type 1 water (18.2 MΩ·cm) under controlled trace-metal and total organic carbon limits, supporting consistent lot-to-lot performance in sensitive lymphocyte cultures.
Low background for imaging
0.04 µm filtration reduces particulate background for flow cytometry and confocal microscopy. This formulation contains phenol red (5.3 mg/L); a phenol red-free variant is available on request for autofluorescence-sensitive PE-channel and biosensor applications.
Rich, stable nutrient profile
RPMI 1640 base with reduced glutathione, 20 amino acids and 11 vitamins, released on a per-lot, micro-batch basis with a full Certificate of Analysis.
Customization on demand
pH, HEPES concentration, salts, and nutrient composition available on request. Contact support@diagnocine.com.
Quadruple-stage filtration system
Four serial filtration stages — two dedicated prefilter + final-filter pairs — reaching a final 0.04 µm polish.
-
1
0.1 µm Prefiltration I
Removes large aggregates and cell debris; protects the first 0.04 µm cartridge.
-
2
0.04 µm Final filtration I
First 0.04 µm pass; retains sub-micron particulates that pass a 0.22 µm filter.
-
3
0.1 µm Prefiltration II
Second dedicated prefilter, protecting the second 0.04 µm cartridge.
-
4
0.04 µm Final filtration II — Polish
Ultimate polishing filter; aseptic fill in an ISO Class 5 (Class 100) environment.
Performance vs. conventional media
A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration, with 0.1 µm mycoplasma-retentive filtration built into two points of the train.

Leukemia & Lymphoma Lines
RPMI supports NCI-60 cancer lines, Jurkat, Raji, K562, HL-60, and other hematopoietic cancer lines where DMEM would alter proliferation and signaling.
Immune Cell OoC
0.04 µm filtered RPMI for tumor-immune interaction chips, vascular-immune OoC, and lymph-node-on-chip models.
CAR-T & TIL Expansion
Low endotoxin release specification (< 0.05 EU/mL) supports CAR-T manufacturing and TIL expansion protocols sensitive to TLR4 activation.
Immune Cell Metabolic Flux
Defined RPMI 1640 + HEPES base with researcher-controlled pyruvate addition, suited to 13C isotope tracing and glycolysis flux studies of lymphocyte activation states. Not compatible with Agilent Seahorse XF assays, which require bicarbonate-free, phenol red-free medium.
Flow Cytometry & Confocal
Ultra-low particulate media suited to flow cytometry and confocal imaging of immune cells. This formulation contains phenol red; ask about our phenol red-free variant for autofluorescence-sensitive PE-channel and biosensor applications.
