 FluxMPS™ RPMI 1640 Medium with 25mM HEPES

Contains L-Glutamine Contains Sodium Bicarbonate Contains Phenol Red Contains HEPES (25 mM) Contains Calcium Contains Magnesium Contains Glucose (2 g/L) Contains Sodium Pyruvate

FluxMPS™ DCP-RPMIH1X is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) RPMI 1640 + 25 mM HEPES formulation engineered for hematopoietic and lymphocyte cell models on organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. The train reaches a 0.04 µm final cut-off — five times finer than the 0.22 µm membranes used in conventional sterile filtration. Dual-buffered with 25 mM HEPES (pKa 7.3 at 37°C) and sodium bicarbonate, it maintains stable pH during both incubator culture and open-air handling. Formulation: [+] 2 g/L Glucose, [+] L-Glutamine, [+] Sodium Pyruvate, [+] Sodium Bicarbonate, [+] 25 mM HEPES.

  • Glucose 2000 mg/L (2.0 g/L) supports high-glycolytic hematopoietic and lymphocyte cell lines
  • 25 mM HEPES (pKa 7.3 at 37°C) plus sodium bicarbonate — dual buffering stable outside CO₂ incubators
  • Glutathione (reduced), 1.000 mg/L — antioxidant support for redox-sensitive lymphocyte cultures
  • Quadruple-stage filtration train (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final cut-off
  • Endotoxin release specification < 0.05 EU/mL, LAL assay (USP <85>)
  • Manufactured under an ISO 13485:2016 quality management system; final QC at Diagnocine, Totowa, NJ
  • 42 formulation components across inorganic salts, amino acids, vitamins and other constituents, released per lot
  • Custom pH, HEPES concentration, and nutrient modifications available on request

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm–filtered RPMI passes mycoplasma-sized particles, subvisible particulates, and endotoxin fragments that activate TLR4 and confound immune cell assays. FluxMPS™ is built to control these failure modes at the filtration stage.

Purity Architecture

Quadruple-stage filtration system

Four serial filtration stages — two dedicated 0.1 µm prefilter + 0.04 µm final-filter pairs — reaching a final 0.04 µm cut-off, five times finer than the 0.22 µm membranes used in conventional sterile filtration.

  1. 1

    0.1 µm Prefiltration I

Removes large particulate, cell debris and protein aggregates; protects the first 0.04 µm final-filter cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass a conventional 0.22 µm filter, including the 0.2–0.3 µm mycoplasma size range.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm cartridge for full-redundancy processing.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter prior to aseptic fill & finish.

Performance vs. conventional media

A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

Sterility & Mycoplasma: No growth after 14-day incubation (USP <71>). Mycoplasma risk is mitigated by 0.1 µm mycoplasma-retentive filtration (not tested per lot); mycoplasma organisms are typically 0.2–0.3 µm in diameter.
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. For applications requiring the 0.01 µm cut-off, contact support@diagnocine.com.

FAQ

Frequently asked questions

Common questions about FluxMPS™ DCP-RPMIH1X — RPMI 1640 + 25 mM HEPES.

Yes. DCP-RPMIH1X is Microfluidics Suitable, processed through a quadruple-stage filtration system (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final cut-off. RPMI 1640 with this filtration profile is suited to immune cell OoC, tumor-immune interaction chips, and lymphocyte perfusion models.
FluxMPS™ uses four sequential filters — 0.1 µm prefiltration I, 0.04 µm final filtration I, 0.1 µm prefiltration II, and 0.04 µm final filtration II (polish) — reaching a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used in conventional sterile filtration.
DCP-RPMIH1X is dual-buffered: 2000 mg/L sodium bicarbonate provides the primary buffering system (5% CO₂ recommended for optimal pH), while 25 mM HEPES (pKa 7.3 at 37°C) adds stability during open-air handling — flow cytometry preparation, assay plate setup, and multi-well stimulation experiments — where CO₂ control is inconsistent.
5% CO₂ recommended. The 2000 mg/L sodium bicarbonate buffering system is calibrated for a 5% CO₂ atmosphere; the added 25 mM HEPES provides supplementary buffering during brief periods outside the incubator.
Yes. FBS (5–10%), serum-free supplements, growth factors, or antibiotics can be added. Serum and other protein-containing supplements should be pre-filtered through a 0.2 µm low-protein-binding PES or PVDF membrane before addition — never through a 0.04 µm membrane, which will retain serum proteins and lipoproteins. Contact support@diagnocine.com for custom co-formulation.
The release specification is < 0.05 EU/mL, verified by LAL assay per USP <85> on every manufacturing batch (assay sensitivity 0.005 EU/mL). This is relevant for immune cell assays, where endotoxin can activate TLR4/NF-κB signaling and alter lymphocyte activation state independently of experimental conditions.
Yes. Full CoA per batch covers: appearance, pH (USP <791>), osmolality (USP <785>), sterility (USP <71>), endotoxin (USP <85>), mycoplasma filtration status, particulate count (USP <788> Method 1), and raw-material traceability. Request at support@diagnocine.com.
