Description:

Potato Dextrose Agar (PDA) plates with ampicillin are a selective fungal medium intended to support robust growth of yeasts and molds while inhibiting many co-present bacteria through the action of a beta-actam antibiotic.  PDA consists of potato infusion/extract, dextrose, and agar, creating a carbohydrate-rich, slightly acidic environment that promotes luxuriant mycelial growth, sporulation, and pigment production in a broad range of environmental, food, and cosmetic yeasts and molds.

Ampicillin is a beta-lactam antibiotic that targets bacterial cell wall synthesis by binding penicillin-binding proteins, leading to lysis in susceptible bacteria, especially many gram-positive and some gram-negative species.  When ampicillin is incorporated into PDA (commonly on the order of tens of µg/mL), it suppresses growth of susceptible bacteria, reducing bacterial background without significantly affecting most filamentous fungi and yeasts, which lack peptidoglycan cell walls.  The intrinsic properties of PDA (nutrient profile and mildly acidic pH) already favor fungal growth over many bacteria; adding ampicillin further decreases bacterial colony formation on the plate, making fungal colonies easier to detect, count, and subculture from mixed samples.
