FluxMPS™ Leibovitz's L-15 Medium: 1X Liquid

Contains L-Glutamine Contains Phenol Red Contains Calcium Contains Magnesium Contains Sodium Pyruvate

Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) 1X liquid Leibovitz's L-15 medium engineered for microfluidic channels, organ-on-a-chip (OoC), and microphysiological systems (MPS). This CO2-independent, bicarbonate-free formulation uses galactose in place of glucose and is manufactured under an ISO 13485:2016 quality management system. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

  • Quadruple-stage filtration (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final pore size for microchannel-safe delivery
  • Galactose-based carbohydrate source (0.9 g/L) replaces glucose, avoiding lactic acid buildup during CO2-independent culture
  • Bicarbonate-free, CO2-independent formulation — no CO2 incubator required
  • L-Glutamine (300 mg/L) and Sodium Pyruvate (550 mg/L) included as ready energy and nitrogen substrates
  • Endotoxin release specification: < 0.05 EU/mL (LAL, USP <85> BET), tested per manufacturing batch
  • Manufactured under an ISO 13485:2016 quality management system; final QC and packaging at Diagnocine, Totowa, NJ
  • Phenol red (11 mg/L) included as a visual pH indicator
  • Custom pH, HEPES, salts & nutrient composition available on request — support@diagnocine.com

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm filtered media allows mycoplasma (0.2–0.3 µm), sub-visible particulates, and microaggregates to pass freely — clogging microchannels and complicating long-duration perfusion. FluxMPS™ closes that gap.

Purity Architecture

Quadruple-stage filtration system

A validated four-stage sequential filtration train reaching a final pore size of 0.04 µm — capturing particulates and organisms that pass through conventional 0.22 µm filtration.

  1. 1

    0.1 µm Prefiltration I

Removes large particulates, cell debris, and protein aggregates; protects the first 0.04 µm final filter cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass a 0.22 µm filter, including organisms in the 0.2–0.3 µm mycoplasma size range.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm final filter cartridge.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter; aseptic fill & finish.

Performance vs. conventional media

By USP <788> Method 1 (light obscuration), FluxMPS™ media are formulated for approximately 5× fewer particles ≥10 µm compared with standard 0.22 µm-filtered media — supporting long-duration perfusion without channel occlusion.

Sterility & Mycoplasma: 14-day USP <71> sterility tested per lot. 0.1 µm mycoplasma-retentive filtration (not tested per lot) provides risk mitigation against mycoplasma contamination (typical diameter 0.2–0.3 µm, well above the 0.1 µm retentive pore size).
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. For applications requiring the 0.01 µm cut-off, contact support@diagnocine.com.

FAQ

Frequently asked questions

Common questions about FluxMPS™ Leibovitz's L-15 Medium: 1X Liquid (DCP-LL151X).

Yes. DCP-LL151X is engineered for organ-on-a-chip, microfluidic, and MPS applications. The 0.04 µm final filtration removes mycoplasma-sized organisms and sub-micron particulates that cause microchannel clogging in standard 0.22 µm media, and the CO2-independent formulation suits closed-system chip culture.
Four sequential stages (0.1 µm ×2 + 0.04 µm ×2) retain particulates and organisms in the 0.2–0.3 µm mycoplasma size range that a single 0.22 µm filter cannot — resulting in approximately 5× fewer particles by USP <788> Method 1 (light obscuration).
Leibovitz's L-15 uses galactose (0.9 g/L) instead of glucose because galactose is metabolized more slowly via oxidative pathways, avoiding the lactic acid buildup that would otherwise occur in this bicarbonate-free, CO2-independent formulation. Sodium pyruvate (550 mg/L) and L-glutamine (300 mg/L) are already included as energy and nitrogen substrates. If your cell line requires glucose for glycolytic flux, supplement with D-glucose per your protocol, or contact support@diagnocine.com for a custom formulation.
No. DCP-LL151X contains no sodium bicarbonate and is formulated for CO2-independent incubation at ambient atmosphere. It is suitable for open-bench, closed-chip, and automated culture systems that lack CO2 control.
Yes. DCP-LL151X is a basal medium compatible with FBS, human serum, growth factors, and antibiotics. Pre-filter serum and protein-containing additions through a 0.2 µm low-protein-binding PES or PVDF filter before combining; a 0.1 µm filter is acceptable for defined, protein-free additions only. Do not use a 0.04 µm filter for supplements — it will strip serum of essential high-molecular-weight components.
Endotoxin is controlled per manufacturing batch to a release specification of < 0.05 EU/mL, tested by LAL assay per USP <85> (Bacterial Endotoxins Test); assay sensitivity is 0.005 EU/mL. Every batch is tested before release, and the lot-specific result is reported on the Certificate of Analysis — request at support@diagnocine.com.
Yes — a full CoA per lot includes: lot number, expiry, appearance, pH (USP <791>), osmolality (USP <785>), endotoxin (USP <85>), sterility (USP <71>), mycoplasma control status, particulate count (USP <788>), and cultural response. Email support@diagnocine.com.
