FluxMPS™ Glasgow's Minimum Essential Medium (GMEM), High Glucose: 2X Liquid

Double-strength (2X) GMEM High Glucose — 9.0 g/L glucose in the concentrate, 4.5 g/L at 1X, with no tryptose phosphate broth or NEAA — for soft-agar, plaque-overlay, amino-acid dropout and other defined or semisolid cultures, ultra-filtered through a quadruple-stage 0.1 µm / 0.04 µm filtration system. With a sodium bicarbonate buffer, it is approximately 5× cleaner than conventional 0.22 µm-filtered media by particulate count.

  • 2X concentrate: mix with an equal volume of melted agar or agarose, or another compatible solution, for a final 1X medium
  • No tryptose phosphate broth or NEAA, for a more defined basal medium you supplement yourself
  • Quadruple-stage filtration: 0.1 µm membrane filtered twice, 0.04 µm membrane filtered twice
  • 0.04 µm nano-filtration final polish acts as a mycoplasma barrier

Why FluxMPS™

Engineered where standard media fails

Conventional GMEM is sterile-filtered once at 0.22 µm. That pore size can pass mycoplasma and leaves subvisible particulates that settle in microchannels, foul membranes and hydrogels, and add noise to optical readouts.[11,12] FluxMPS™ GMEM keeps the Glasgow formulation and removes that failure mode.

Applications

Where FluxMPS™ GMEM fits

Glasgow's Minimum Essential Medium (GMEM) is a modified Basal Medium Eagle (BME) formulation.[2] Macpherson and Stoker originally added tryptose phosphate broth and doubled BME's amino acids and vitamins to grow BHK-21 clone 13 cells.[1,3] This version contains no tryptose phosphate broth and no added non-essential amino acids. That makes it a more defined basal medium that researchers can supplement to suit their own cell systems.

GMEM has no proteins, lipids, or growth factors, so it must be supplemented for cell growth. It uses a sodium bicarbonate buffer and needs a 5–10% CO₂ atmosphere to keep physiological pH. Users should check the literature for supplements and growth requirements specific to their cell lines.

GMEM 2X, Without Tryptose Phosphate Broth and NEAA, is a double-strength version of the 1X basal formulation. It's meant to be combined with an equal volume of a compatible solution so the basal ingredients end up at 1X. This is the standard approach for agar and agarose cultures, where 2X medium is mixed 1:1 with melted agar or agarose.[4] Because the formulation contains no TPB or NEAA, researchers can add their own supplements, test compounds, cells, or gelling agents to the 2X medium while keeping the final basal concentration under control. Final pH, osmolality, buffering, supplementation, and gel compatibility have to be confirmed for each research system.

Why omit TPB and NEAA?

  • Without TPB: TPB contains tryptose, a peptone made by enzymatically digesting the milk protein casein. Leaving it out removes an undefined, hydrolysate-derived source of nutrients and survival factors.
  • Without NEAA: Standard MEM NEAA supplement supplies seven amino acids: glycine, L-alanine, L-asparagine, L-aspartic acid, L-glutamic acid, L-proline, and L-serine. Researchers who omit them can add back selected amino acids at controlled levels.

It is useful for:

  • Defined or serum-free medium development
  • Amino-acid dropout and add-back studies
  • Serine/glycine and nutrient-metabolism research
  • BHK-21 and kidney-cell research
  • Stem-cell differentiation research
  • Soft agar and anchorage-independent growth assays
  • Virology plaque or overlay assays
  • Customized research media

Automated Bioreactors & Robotics

Next-Generation System Uptime

For automated bioreactors, liquid-handling robots and closed perfusion loops with fine valves and in-line sensors, an optional 10 nm (0.01 µm) ultra-filtered grade of this medium is available.

  • Total Particulate Exclusion: 0.01 µm filtration removes virtually all particulates down to the nanometer scale.
  • Valve & Sensor Protection: keeps micro-valves, pumps and optical or electrochemical sensors free of deposits.
  • Extended Perfusion Stability: supports long, uninterrupted perfusion runs with fewer line changes.

Note: pH and sodium bicarbonate concentration of the prepared medium are critical factors affecting cell growth. This is also influenced by amount of medium and volume of culture vessel used (surface to volume ratio). For example, in large bottles, such as Roux bottles, pH tends to rise perceptibly as a significant volume of carbon dioxide is released. Therefore, optimal conditions of pH, sodium bicarbonate concentration and surface to volume ratio must be determined for each cell type. We recommend stringent monitoring of pH. If needed, pH can be adjusted by using sterile 1N HCl or 1N NaOH or by bubbling in carbon dioxide. If required, supplements can be added to the medium prior to or after filter sterilization observing sterility precautions.

FAQ

Frequently asked questions

Common questions about using DCP-GMEM-JN2X for semisolid, 3D, microfluidic and conventional culture.

Yes. DCP-GMEM-JN2X is quadruple-stage filtered to a 0.04 µm final pore size, so it carries far fewer subvisible particles than 0.22 µm media — approximately 5× cleaner by particulate count. That lowers the risk of clogging microchannels, valves and membranes in organ-on-a-chip, hydrogel-based 3D and microfluidic perfusion systems.[8,9,10]
The medium is filtered four times: 0.1 µm twice and 0.04 µm twice. A single 0.22 µm pass can let mycoplasma and fine particulates through; the 0.04 µm stages act as a mycoplasma barrier and polish out sub-micron particles before ISO Class 5 aseptic fill.[11]
HEPES is not included, so the medium relies on its bicarbonate buffer in a 5–10% CO₂ atmosphere; choose the 50 mM HEPES version if you need extra buffering outside the incubator. Non-essential amino acids are left out. The standard MEM NEAA supplement supplies seven amino acids — glycine, L-alanine, L-asparagine, L-aspartic acid, L-glutamic acid, L-proline and L-serine — so omitting it lets you add back selected amino acids at controlled levels for dropout, add-back and serine/glycine metabolism studies. Tryptose phosphate broth is left out because tryptose is a peptone made by enzymatically digesting the milk protein casein — an undefined, hydrolysate-derived source of nutrients and survival factors. Macpherson and Stoker's original GMEM included it for BHK-21 clone 13 cells;[1,3] leaving it out gives a more defined basal medium you can supplement yourself. GMEM has no proteins, lipids or growth factors, so it must be supplemented for cell growth. Users are advised to review the literature for supplementation and physiological growth requirements specific to each cell type.[1,2]
Yes. The medium uses a sodium bicarbonate buffer (2,750 mg/L at 1X) and needs a 5–10% CO₂ atmosphere to keep physiological pH; keep time in room air short.
Mix 2X GMEM with an equal volume of a compatible solution or matrix — melted agar or agarose, supplements, test compounds or cells — so glucose (4.5 g/L), amino acids, vitamins and salts end up at their 1X concentration.[4] Final pH, osmolality, buffering and gel compatibility should be confirmed for each research system. Serum, antibiotics and other supplements can be added before or after filter sterilization, observing sterility precautions.
The endotoxin specification is < 0.05 EU/mL, tested by the USP <85> bacterial endotoxins test (BET) and reported on the lot Certificate of Analysis. The medium is made with Ultrapure Type 1 water (18.2 MΩ·cm).
Yes. Each lot ships with a CoA covering appearance (red), pH 7.4 (± 0.04), sterility (no bacterial or fungal growth after 14 days of incubation per USP), endotoxin and osmolality results. Request copies at support@diagnocine.com.
