TLR9 Ligand (CpG-ODN) for pDCs and B-cells D35 and K3
CpG Oligodeoxy nucleotide (CpG-ODN), which contains CpG-motif, has a function to stimulate immune system through Toll-like receptor 9 (TLR9) and promote maturation and activation of antigen presenting cells (APCs). This mimics immuno-stimulation activity of bacterial DNA.
D35 is categorized in "A-class" TLR9 ligand. D35 strongly induces production of type I Interferon (IFN) by activating pDCs and PBMCs, but it shows lower activity to B-cells.
K3 is categorized in "B-class" TLR9 ligand. K3 induces activation of TLR9-dependent NF-kB signal by stimulating B-cells, but it shows lower activity for inducing IFN-alpha.

Human PBMC (1 x 10^6 cells) were stimulated with each CpG (1 μM) for 24 hours.IFN-alpha and IL-6 in culture supernatant were measured by ELISA. Bar graph shows average +/- SEM.

Fig. 2 Cytokine induction from mouse FL-DCs
FL-DCs (2 x 10^6 cells) derived from mouse bone marrow were differentiated by FLT3L and stimulated by each CpG (1 μM) for 24 hours. IFN-alpha and IL-6 in culture supernatant were measured by ELISA. Bar graph shows average +/- SEM.
