Description:
Base Columbia agar with ampicillin sodium salt is a nutrient-rich, non-blood agar medium in which a standard Columbia agar base is supplemented with the β-lactam antibiotic ampicillin (usually as the sodium salt) to give it selective properties against ampicillin-susceptible bacteria. Columbia agar typically contains enzymatic digests of casein and meat, yeast extract, starch, and NaCl. The peptones and yeast extract provide amino acids, peptides, nitrogen, vitamins, and growth factors that support abundant growth of both fastidious and non-fastidious organisms. Starch (corn/potato) acts as a carbon source and can bind toxic metabolites, which helps recovery of delicate organisms, while NaCl maintains osmotic balance and agar provides the solid support for colony growth.
When ampicillin sodium salt is added, the medium becomes selective because ampicillin interferes with bacterial cell wall synthesis. As a β-lactam, ampicillin preferentially inhibits many Gram-positive and some Gram-negative organisms that do not carry β-lactamases or other resistance mechanisms.
