FluxMPS™ RPMI 1640 + 25mM HEPES

Contains Phenol Red Contains 25mM HEPES Contains Calcium Contains Magnesium Contains 2.0 g/L Glucose Contains Sodium Pyruvate Without L-Glutamine Without Sodium Bicarbonate

FluxMPS™ DCP-RPMIH-QB1X is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) RPMI 1640 + 25mM HEPES formulation engineered for hematopoietic cells, lymphocytes and related immune cell models on organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. The quadruple-stage train reaches a 0.04 µm final cut-off — five times finer than the 0.22 µm membranes used for conventional sterile filtration. HEPES (25 mM, pKa 7.3 at 37°C) provides CO2-independent pH buffering; this formulation excludes L-glutamine and sodium bicarbonate so users can control nitrogen source and buffering strategy at time of use.

  • Carbon source: 2.0 g/L (2000 mg/L) D-glucose, plus 110 mg/L sodium pyruvate
  • 25 mM HEPES (pKa 7.3 at 37°C) — CO2-independent pH buffering for open-air handling and flow cytometry preparation
  • Glutathione (reduced), 1.0 mg/L — antioxidant support for redox-sensitive lymphocyte populations
  • Quadruple-stage filtration train: 0.1 µm → 0.04 µm → 0.1 µm → 0.04 µm, reaching a 0.04 µm final cut-off
  • Endotoxin release specification: < 0.05 EU/mL (LAL, USP <85>)
  • L-glutamine and sodium bicarbonate excluded — add fresh glutamine and select a buffering strategy suited to your culture system
  • Manufactured under an ISO 13485:2016 quality management system; final QC at Diagnocine, Totowa, NJ
  • Contains phenol red (5.3 mg/L) — a phenol red-free variant is available on request for fluorescence-sensitive imaging

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm–filtered RPMI passes mycoplasma-sized organisms, subvisible particulates, and endotoxin fragments that can activate TLR4 signaling and confound immune cell assays. FluxMPS™ is built to mitigate these failure modes at the filtration stage.

Purity Architecture

Quadruple-stage filtration system

Four serial filtration stages — two dedicated prefilter/final-filter pairs — reaching a 0.04 µm final pore size under ISO Class 5 aseptic conditions.

  1. 1

    0.1 µm Prefiltration I

Removes large particulate, cell debris and protein aggregates; protects the first 0.04 µm cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates not captured by standard 0.22 µm membranes.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm cartridge and providing 0.1 µm mycoplasma-retentive filtration.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter; aseptic fill and finish performed in validated ISO Class 5 (Class 100) laminar-flow workstations.

Performance vs. conventional media

FluxMPS™ DCP-RPMIH-QB1X is processed through a validated quadruple-stage filtration train reaching a 0.04 µm final cut-off — five times finer than the 0.22 µm membranes used in conventional filtration — with 0.1 µm mycoplasma-retentive prefiltration at two points in the process.

5×
Finer final cut-off than 0.22 µm conventional media (0.04 µm vs 0.22 µm)
4
Sequential filtration passes (0.1 µm ×2 + 0.04 µm ×2)
Sterility & Mycoplasma: No growth after 14-day incubation (USP <71>). Mycoplasma risk (organisms 0.2-0.3 µm in diameter) is mitigated through 0.1 µm mycoplasma-retentive filtration; mycoplasma testing is not performed on every lot.
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. FAQ

Frequently asked questions

Common questions about FluxMPS™ DCP-RPMIH-QB1X — RPMI 1640 + 25mM HEPES.

Yes. DCP-RPMIH-QB1X is processed through a quadruple-stage filtration system reaching a 0.04 µm final pore size, making it Microfluidics Suitable for MPS, OoC, and LoC platforms. RPMI 1640 with 0.1 µm mycoplasma-retentive filtration is suited to immune cell OoC, tumor-immune interaction chips, and lymphocyte perfusion models.
FluxMPS™ uses four sequential filters — 0.1 µm prefiltration I, 0.04 µm final filtration I, 0.1 µm prefiltration II, and 0.04 µm final filtration II (polish) — reaching a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used in conventional filtration, with 0.1 µm mycoplasma-retentive prefiltration at two points in the process.
This formulation is HEPES-buffered and bicarbonate-free by design, so buffering is CO2-independent. L-glutamine is left out because it degrades in liquid storage; add fresh L-glutamine (commonly 2 mM) or a stabilized dipeptide substitute at time of use, tailored to your cell type's nitrogen requirements.
No. This formulation is CO2-independent — 25 mM HEPES alone maintains pH 7.2-7.4 at 37°C, making it suitable for atmospheric handling and open-bench workflows.
Yes. Add FBS (typically 5-10%), serum-free supplements, growth factors, or antibiotics as required. When adding serum or protein-containing supplements, use a 0.2 µm low-protein-binding PES or PVDF filter (never 0.04 µm, which retains IgM, VLDL, and much of the lipid/lipoprotein fraction of serum). Contact support@diagnocine.com for custom co-formulation.
Every batch is tested by LAL assay (USP <85>, assay sensitivity 0.005 EU/mL) and must meet the release specification of < 0.05 EU/mL before it ships. Endotoxin is controlled per manufacturing batch rather than per unit. For immune cell assays, endotoxin above typical thresholds can activate TLR4/NF-κB, inducing cytokines and altering lymphocyte activation state independently of experimental conditions.
Yes. A full CoA per lot covers appearance, pH (USP <791>), osmolality (USP <785>), sterility (USP <71>), endotoxin (USP <85>), mycoplasma filtration status, particulate count (USP <788> Method 1), and raw-material traceability. Request at support@diagnocine.com.
