FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate, Phenol Red: 1X Liquid

Contains Calcium Contains Magnesium Contains Glucose (4.5 g/L) Contains Sodium Pyruvate Without L-Glutamine Without Sodium Bicarbonate Without Phenol Red Without HEPES

FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate, Phenol Red: 1X Liquid is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) 1X liquid cell culture medium engineered for organ-on-a-chip (OoC), tissue-on-a-chip (ToC), and microphysiological system (MPS) applications. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

  • Quadruple-stage filtration: 0.1 µm (Prefiltration I & II) + 0.04 µm (Final filtration I & II), reaching a 0.04 µm final cut-off
  • Endotoxin release specification: < 0.05 EU/mL (LAL assay, USP <85> BET), tested per manufacturing batch
  • RPMI 1640 high-glucose base (4.5 g/L D-glucose) with 110 mg/L sodium pyruvate, calcium and magnesium salts
  • Formulated without L-glutamine, sodium bicarbonate, phenol red, and HEPES for downstream customization
  • pH 7.4 (USP <791>); bicarbonate-free and HEPES-free — buffering system added by the end user
  • Manufactured under an ISO 13485:2016 quality management system; final packaging and testing at Diagnocine, Totowa, NJ
  • Custom formulations available — pH, glucose, salts, HEPES, and nutrient composition on request

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm-filtered media carry mycoplasma-sized particulates (0.2–0.3 µm), subvisible debris, and endotoxin variability that accumulate inside microchannels — corrupting biosensor readings, triggering inflammation in primary cultures, and shortening device lifetimes. FluxMPS™ is built to reduce these failure modes at the filtration stage.

filter_alt

Microchannel-safe purity

0.04 µm final filter targets particles down to sub-mycoplasma size; USP <788> Method 1 (light obscuration) particulate compliance verified per lot.

target

Total metabolic control

Defined glucose (4.5 g/L) and sodium pyruvate (110 mg/L) concentrations, without L-glutamine or bicarbonate, support user-controlled metabolic flux and carbon-source experiments.

water_drop

Ultrapure-grade water

Prepared with Type 1 water (18.2 MΩ·cm, ASTM D1193 Type I), supporting a clean, low trace-metal formulation base.

visibility

Low background for imaging

Ultra-low particulate baseline supports high-content confocal microscopy, live-cell biosensors, and TEER measurements. Note: this formulation contains riboflavin, which carries intrinsic autofluorescence — validate imaging channels accordingly.

Purity Architecture

Quadruple-stage filtration system

FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate, Phenol Red: 1X Liquid is processed through a four-stage serial filtration train — two dedicated 0.1 µm/0.04 µm prefilter-and-final-filter pairs in series — reaching a 0.04 µm final pore size.

  1. 1

    0.1 µm Prefiltration I

Removes large particulates and protein aggregates; protects the first 0.04 µm cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass a 0.22 µm filter, including mycoplasma-sized particles (0.2–0.3 µm).

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm cartridge from breakthrough particulate.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter ahead of aseptic fill in an ISO Class 5 laminar-flow workstation.

Performance vs. conventional media

Four sequential stages reaching 0.04 µm deliver approximately 5× cleaner media by particulate count compared to single-pass 0.22 µm filtration.

Sterility assurance: Every lot undergoes 14-day USP <71> sterility testing. Mycoplasma risk is controlled by 0.1 µm mycoplasma-retentive filtration (not tested per lot) — this is a filtration control, not a mycoplasma test result.
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. FAQ

Frequently asked questions

Common questions about FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate, Phenol Red: 1X Liquid.

Yes. DCP-RPMIG-QBR1X uses a quadruple-stage filtration train (0.1 µm ×2 + 0.04 µm ×2), delivering ultra-low particulate counts that reduce microchannel clogging risk in OoC and MPS devices.
Standard 0.22 µm filtration leaves intact mycoplasma-sized particles (0.2–0.3 µm) and subvisible particulates that accumulate in microchannels. FluxMPS™ uses four sequential stages reaching 0.04 µm, delivering approximately 5× lower particulate counts per USP <788> Method 1.
This formulation is supplied without L-glutamine, sodium bicarbonate, and phenol red so users can add their preferred buffering system, glutamine source, and pH indicator based on their assay. Add L-glutamine or a stable dipeptide substitute, sodium bicarbonate or HEPES for buffering, and phenol red only if visual pH monitoring is required. Contact support@diagnocine.com for a custom pre-formulated version.
This formulation contains no sodium bicarbonate and no HEPES. CO₂ requirement depends entirely on the buffering system you add before use: adding sodium bicarbonate typically requires a matched CO₂ incubator atmosphere, while adding HEPES allows ambient-CO₂ culture. Contact support@diagnocine.com for buffering guidance specific to your application.
Yes. FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate, Phenol Red: 1X Liquid can be supplemented with FBS (5–20%), growth factors, antibiotics, or other additives per standard practice. When adding serum or protein-containing supplements, filter through a 0.2 µm low-protein-binding PES or PVDF membrane — never 0.04 µm, which retains serum proteins and lipoproteins. Add supplements immediately before use.
Endotoxin is controlled per manufacturing batch, not per unit. Every batch is tested by LAL assay per USP <85> BET (assay sensitivity 0.005 EU/mL) and must meet the release specification of < 0.05 EU/mL before shipment. Batch-specific results are documented in the CoA, available from support@diagnocine.com.
Yes. A lot-specific CoA is available for every shipment and includes: appearance, pH (USP <791>), osmolality (USP <785>), endotoxin (USP <85> BET), sterility (USP <71>), particulate matter (USP <788> Method 1), raw material traceability, lot number, expiry, manufacturing date, and authorized release signatures.
