FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate: 1X Liquid

Contains Phenol Red Contains Calcium Contains Magnesium Contains Glucose Contains Sodium Pyruvate Without L-Glutamine Without Sodium Bicarbonate

FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate: 1X Liquid is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) 1X liquid cell culture medium engineered for organ-on-a-chip (OoC), tissue-on-a-chip (ToC), and microphysiological system (MPS) applications. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

  • Quadruple-stage filtration: 0.1 µm (Prefiltration I & II) + 0.04 µm (Final filtration I & II — Polish), reaching a 0.04 µm final cut-off
  • Endotoxin release specification: < 0.05 EU/mL (LAL assay, USP <85> BET), tested per manufacturing batch
  • RPMI 1640 high-glucose base (4500 mg/L / 4.5 g/L) formulated without L-glutamine and without sodium bicarbonate — buffering and glutamine to be added at time of use
  • Contains 110 mg/L sodium pyruvate and phenol red sodium salt (5.3 mg/L) as a visual pH indicator
  • Prepared with Ultrapure Type 1 water (18.2 MΩ·cm); ISO Class 5 (Class 100) aseptic fill & finish
  • Manufactured under an ISO 13485:2016 quality management system; final QC and packaging at Diagnocine, Totowa, NJ
  • Custom formulations available — pH, glucose, salts, HEPES, and nutrient composition on request

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm-filtered media carry mycoplasma-sized particulates (0.2–0.3 µm), subvisible debris, and endotoxin variability that accumulate inside microchannels — corrupting biosensor readings and shortening device lifetimes. FluxMPS™ is built to reduce these failure modes at the filtration stage.

Purity Architecture

Quadruple-stage filtration system

FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate: 1X Liquid is processed through a four-pass, paired prefilter + final-filter sequence reaching a 0.04 µm final cut-off — addressing mycoplasma-sized particulate and subvisible debris that a single 0.22 µm pass does not remove.

  1. 1

    0.1 µm Prefiltration I

Removes large particulates and protein aggregates; protects the first 0.04 µm final filter cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass a 0.22 µm filter.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm final filter cartridge.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter; aseptic fill & finish in a validated ISO Class 5 laminar-flow workstation.

Performance vs. conventional media

Four sequential passes reaching 0.04 µm deliver approximately 5× cleaner media by particulate count compared to single-pass 0.22 µm filtration, with USP <788> Method 1 compliance verified on every lot.

Sterility assurance: Every batch undergoes 14-day USP <71> sterility testing. Mycoplasma risk is mitigated by 0.1 µm mycoplasma-retentive filtration; lots are not individually tested to USP <63>.
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. For applications requiring the 0.01 µm cut-off, contact support@diagnocine.com.

FAQ

Frequently asked questions

Common questions about FluxMPS™ RPMI 1640 Medium, High Glucose w/o L-Glutamine, Sodium Bicarbonate: 1X Liquid and Microfluidics Suitable cell culture media.

Yes. DCP-RPMIG-QB1X is processed through Quadruple-stage filtration (0.1 µm ×2 + 0.04 µm ×2), delivering an ultra-low particulate baseline that supports microchannel patency in OoC and MPS devices.
Standard 0.22 µm filtration leaves intact mycoplasma-sized particulate (0.2–0.3 µm) and subvisible debris that accumulate in microchannels. FluxMPS™ runs four sequential passes reaching 0.04 µm, delivering approximately 5× lower particulate counts per USP <788> Method 1.
This formulation is supplied without L-glutamine (which is unstable in liquid storage and typically added fresh, or as a stabilized dipeptide, at time of use) and without sodium bicarbonate, so the buffering system can be matched to your culture setup. Add L-glutamine (typically 2 mM) and either sodium bicarbonate (for CO₂ incubator use) or HEPES (for ambient buffering) per your protocol. Contact support@diagnocine.com for a custom pre-supplemented formulation.
This formulation contains neither sodium bicarbonate nor HEPES buffer. It is not pre-buffered for a specific atmosphere; supplement with sodium bicarbonate for CO₂ incubator use, or with HEPES for ambient (non-CO₂) culture, based on your protocol.
Yes. This medium can be supplemented with FBS (5–20%), growth factors, antibiotics, or other additives per standard practice. Add supplements immediately before use. Filter serum-containing or protein-containing supplements through a 0.2 µm low-protein-binding PES or PVDF membrane before addition; use 0.1 µm only for defined, protein-free additions. Do not use a 0.04 µm membrane for supplement filtration — it retains serum proteins and lipoproteins and clogs rapidly.
Endotoxin is controlled per manufacturing batch by LAL assay per USP <85> Bacterial Endotoxins Test (assay sensitivity 0.005 EU/mL). Every batch is tested before release and must meet the specification: < 0.05 EU/mL. Lot-specific results are documented in the Certificate of Analysis, available from support@diagnocine.com.
Yes. A lot-specific CoA is available for every shipment and includes: appearance, pH (USP <791>), osmolality (USP <785>), endotoxin (USP <85> BET), sterility (USP <71>), particulate matter (USP <788> Method 1), raw material traceability, manufacturing date, lot number, expiry, and authorized release signatures.
