FluxMPS™ Leibovitz's L-15 Medium with 50mM HEPES — 2X Liquid

Contains HEPES (50 mM) Contains Calcium Contains Magnesium Without L-Glutamine Without Phenol Red Without Sodium Pyruvate

FluxMPS™ DCP-LL15H-QPR2X is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) Leibovitz's L-15 2X + 50 mM HEPES formulation engineered for primary cells and related models on microfluidic, organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration. HEPES (50 mM, pKa 7.3 at 37°C) and a D-galactose carbon source provide CO2-independent pH stability for open-stage and atmospheric culture work.

  • CO2-free by design: 1.8 g/L D-galactose energy source plus high amino acid buffering — no gas supplementation required
  • 50 mM HEPES (pKa 7.3 at 37°C) provides robust pH buffering independent of CO2
  • Quadruple-stage filtration system (0.1 µm ×2 + 0.04 µm ×2) to a 0.04 µm final cut-off
  • Endotoxin release specification: < 0.05 EU/mL, LAL assay per USP 85 (Bacterial Endotoxins Test)
  • Formulated without L-glutamine, sodium pyruvate, or phenol red for full researcher-defined supplementation
  • Manufactured under an ISO 13485:2016 quality management system; final QA at Diagnocine, Totowa, NJ
  • Ultrapure Type 1 water (18.2 MΩ·cm) and ISO Class 5 (Class 100) aseptic fill

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm–filtered Leibovitz's L-15 passes mycoplasma-sized organisms and subvisible particulates that interfere with CO2-free pH maintenance, microchannel integrity, and live-cell imaging clarity. FluxMPS™ is built to remove those failure modes.

filter_alt

Microchannel-safe purity

0.04 µm final filtration and USP <788> particulate compliance support clean live imaging and CO2-free chip loading.

science

CO2-free by design formulation

Leibovitz's L-15 uses D-galactose (not glucose) as the primary carbon source, together with high amino acid concentrations, for CO2-free pH buffering. This is a 2X concentrated formulation; 50 mM HEPES adds further buffering capacity for prolonged open-stage procedures.

water_drop

Ultrapure-grade water

18.2 MΩ·cm Type 1 water, controlled for trace metals and organic carbon, supports the amino acid buffering system that gives L-15 its CO2-free stability.

shield

Low endotoxin specification

< 0.05 EU/mL release specification. Endotoxin (LPS) is a known TLR4 agonist; the specification limits the potential for inflammatory activation of primary cell models.

water_drop

HEPES pH stability

50 mM HEPES resists pH rise during open-air handling, flow cytometry preparation, and atmospheric incubation.

Purity Architecture

Quadruple-stage filtration system

Four serial filtration stages, run as two dedicated prefilter + final-filter pairs, reach a final 0.04 µm polish under aseptic conditions.

  1. 1

    0.1 µm Prefiltration I

Large particulate, cell debris and protein aggregate removal; 0.1 µm mycoplasma-retentive filtration (not tested per lot); protects the first 0.04 µm cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass a 0.22 µm filter.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm cartridge.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter; ISO Class 5 (Class 100) aseptic fill & finish.

Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. FAQ

Frequently asked questions

Common questions about FluxMPS™ DCP-LL15H-QPR2X — Leibovitz's L-15 2X + 50 mM HEPES.

Yes. DCP-LL15H-QPR2X is processed through a quadruple-stage filtration system reaching a 0.04 µm final pore size, delivering low particulate levels suited to microfluidic, OoC, and MPS platforms. Its CO2-free design makes it well suited to chip loading, priming, and open-stage imaging without CO2 incubation.

No phenol red, reducing absorbance and fluorescence interference in colorimetric and imaging-based assays; no L-glutamine or sodium pyruvate, allowing full researcher-defined supplementation. Add L-glutamine or a stabilized dipeptide substitute, sodium pyruvate, and/or phenol red per your assay's requirements at the time of use.
No. This formulation is CO2-independent by design: D-galactose and high amino acid concentrations maintain pH without gas supplementation, making it suitable for open-air handling and atmospheric incubation.
Yes. Add FBS (5–10%), serum-free supplements, growth factors, or antibiotics as required. When filtering serum-containing supplements post-addition, use a 0.2 µm low-protein-binding PES or PVDF filter — never 0.04 µm, which retains serum proteins and lipoproteins. Contact support@diagnocine.com for custom co-formulation.
FluxMPS™ DCP-LL15H-QPR2X is released to a batch specification of < 0.05 EU/mL by LAL assay (USP <85>), controlled per manufacturing batch rather than per unit. Elevated endotoxin (LPS) is a known TLR4 agonist that can alter primary cell phenotype and differentiation, which is why the specification is set at this level.
Yes. Full CoA per batch covers: appearance, pH (USP <791>), osmolality (USP <785>), sterility (USP <71>), endotoxin (USP <85>), mycoplasma control statement, particulate count (USP <788> Method 1), lot number, expiry, and raw-material traceability. Request at support@diagnocine.com.
