Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm-filtered RPMI passes subvisible particulates, mycoplasma in the 0.2–0.3 µm range, and endotoxin fragments that can confound sensitive lymphocyte and immune-cell assays. FluxMPS™ RPMI 1640 + 25mM HEPES is built to reduce these variables.[1]

filter_alt

Microchannel-safe purity

0.04 µm final filtration and USP <788> Method 1 (light obscuration) particulate compliance provide particulate-clean media for suspension immune cell OoC, flow cytometry, and lymphocyte perfusion chips.

target

Total metabolic control

L-glutamine, sodium pyruvate, and sodium bicarbonate are withheld so glucose (2.0 g/L), amino acids, and buffering can each be titrated independently for immune-cell metabolic flux and Warburg-effect studies under HEPES-buffered, CO2-independent conditions.

water_drop

Ultrapure-grade water

Type 1 water (18.2 MΩ·cm) feed stock supports low trace-metal and organic-carbon (TOC) background for sensitive lymphocyte and hematopoietic cell culture.

visibility

Low background for imaging

Quadruple-stage 0.04 µm filtration reduces particulate background for confocal imaging, biosensor, and flow cytometry channels. This formulation contains phenol red sodium salt (5.3 mg/L); a phenol-red-free variant is available on request for fluorescence-sensitive assays.

science

Rich, stable nutrient profile

RPMI 1640's amino acid and vitamin base, including reduced glutathione (1.0 mg/L) for redox support, is manufactured in micro-batches with full lot-to-lot CAS traceability across all 39 formulation components.

Purity Architecture

Quadruple-stage filtration system

Four serial filtration stages — two dedicated 0.1 µm prefilter / 0.04 µm final-filter pairs — reach a 0.04 µm final cut-off prior to aseptic fill.[4]

  1. 1

    0.1 µm Prefiltration I

Removes large aggregates and cell debris; protects the first 0.04 µm final-filter cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass a conventional 0.22 µm filter, including the 0.2–0.3 µm mycoplasma size range.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm final-filter cartridge.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter prior to aseptic fill & finish.

Performance vs. conventional media

A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

Sterility & Mycoplasma: No growth after 14-day incubation (USP <71>). Mycoplasma is controlled via 0.1 µm and 0.04 µm mycoplasma-retentive filtration at every stage (not tested per lot; typical mycoplasma diameter 0.2–0.3 µm).
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. For applications requiring the 0.01 µm cut-off, contact support@diagnocine.com.

FAQ

Frequently asked questions

Common questions about FluxMPS™ DCP-RPMIH-QPB1X — RPMI 1640 + 25mM HEPES.

Yes. DCP-RPMIH-QPB1X is processed through a quadruple-stage filtration system (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final pore size, making it Microfluidics Suitable for immune-cell OoC, tumor-immune interaction chips, and lymphocyte perfusion models on microphysiological system (MPS) platforms.
FluxMPS™ uses four sequential filters — 0.1 µm Prefiltration I, 0.04 µm Final filtration I, 0.1 µm Prefiltration II, and 0.04 µm Final filtration II — reaching a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used in conventional sterile filtration.
This HEPES-only, bicarbonate-free formulation lets glutamine, pyruvate, and buffering be titrated independently of the base medium, giving maximum flexibility for immune-cell metabolic flux analysis in CO2-independent conditions. Add L-glutamine or a stable dipeptide substitute, sodium pyruvate, and/or sodium bicarbonate as required for your specific cell line and assay format. Contact support@diagnocine.com for a pre-supplemented custom formulation.
No. This formulation is bicarbonate-free and HEPES-buffered (25 mM, pKa 7.3 at 37°C), providing CO2-independent pH stability for open-air handling and non-CO2 incubation. Validate pH stability under your specific culture conditions and cell line.
Yes. FBS (typically 5–10%), serum-free supplements, growth factors, or antibiotics can be added as required. Filter serum-containing or protein-containing additions through a 0.2 µm low-protein-binding PES or PVDF membrane; use 0.1 µm only for defined, protein-free additions. Never filter serum through 0.04 µm — that pore size retains IgM, VLDL, chylomicrons, and much of the serum lipoprotein fraction. Contact support@diagnocine.com for custom co-formulation.
Endotoxin is controlled per manufacturing batch rather than per unit. Every batch is tested by LAL assay (USP <85>, assay sensitivity 0.005 EU/mL) and must meet the release specification of < 0.05 EU/mL before release. Low endotoxin levels reduce the risk of LPS-driven TLR4/NF-κB activation, which can confound lymphocyte proliferation and cytokine assays.
Yes. Full CoA per lot covers: appearance, pH (USP <791>), osmolality (USP <785>), sterility (USP <71>), endotoxin (USP <85>), mycoplasma filtration status, particulate count (USP <788> Method 1), lot number, expiry, and raw-material traceability. Request at support@diagnocine.com.
