FluxMPS™ RPMI 1640 Medium with 25mM HEPES w/o L-Glutamine: 1X Liquid
FluxMPS™ DCP-RPMIH-Q1X is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) RPMI 1640 + 25 mM HEPES formulation engineered for hematopoietic and lymphoid cell models on organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. The 0.04 µm final cut-off is five times finer than the 0.22 µm membranes used for conventional sterile filtration. HEPES (25 mM, pKa 7.3 at 37°C) supplements the sodium bicarbonate buffering system for added pH stability. Formulation: [+] 2.0 g/L Glucose, [+] Sodium Pyruvate, [+] Sodium Bicarbonate, [+] 25mM HEPES, [+] Phenol Red | [-] L-Glutamine (add fresh at time of use).
- Glucose (carbon source): 2000 mg/L (2.0 g/L), matching classical RPMI 1640
- 25 mM HEPES (pKa 7.3 at 37°C) buffering, supplementing the 2000 mg/L sodium bicarbonate system
- Reduced glutathione (1.0 mg/L) included — antioxidant support for redox-sensitive lymphoid cell types
- L-Glutamine omitted for fresh addition at time of use, avoiding in-bottle degradation and ammonia accumulation
- Quadruple-stage filtration train (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final cut-off; endotoxin release specification < 0.05 EU/mL (LAL, USP <85>)

Engineered where standard media fails
Conventional 0.22 µm–filtered RPMI passes mycoplasma-sized organisms, subvisible particulates, and endotoxin fragments that can activate TLR4 signaling and confound sensitive immune-cell assays. FluxMPS™ is built to reduce those failure modes.
Microchannel-safe purity
A 0.04 µm final filtration stage and USP <788> Method 1 (light obscuration) particulate compliance support particle-sensitive suspension immune-cell OoC and flow cytometry workflows.
Immune cell–optimized formulation
RPMI 1640 provides reduced glutathione (antioxidant) and a balanced amino acid/vitamin profile suited to lymphocyte and hematopoietic cell culture.
Ultrapure-grade water
Formulated with Type 1 water (18.2 MΩ·cm), minimizing trace ionic and organic contaminants relevant to sensitive lymphocyte signaling assays.
Low endotoxin release specification
Released at < 0.05 EU/mL by LAL assay (USP <85>), reducing the risk of LPS-driven TLR4 activation artefacts in immune-cell assays.
HEPES pH stability
25 mM HEPES (pKa 7.3 at 37°C) supplements the sodium bicarbonate buffering system, helping stabilize pH during open-bench handling and flow cytometry preparation.

Quadruple-stage filtration system
Four sequential filtration passes — two dedicated prefilter/final-filter pairs — reach a 0.04 µm final cut-off under aseptic conditions.
-
1
0.1 µm Prefiltration I
Removes large aggregates and cellular debris; protects the downstream 0.04 µm final filter cartridges.
-
2
0.04 µm Final filtration I
First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass through a conventional 0.22 µm filter, and provides mycoplasma-retentive margin (mycoplasma organisms are typically 0.2–0.3 µm in diameter).
-
3
0.1 µm Prefiltration II
A second, dedicated 0.1 µm prefilter protects the second 0.04 µm cartridge, giving the train full redundancy.
-
4
0.04 µm Final filtration II — Polish
Ultimate polishing filter; aseptic fill & finish under ISO Class 5 (Class 100) conditions.
Performance vs. conventional media
A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

T Cell & Lymphocyte Culture
RPMI 1640 is the standard base for primary T cells, B cells, NK cells, and monocytes. FluxMPS™ 0.04 µm filtration helps reduce particulate-driven TLR4 activation that can confound immune assays.
Leukemia & Lymphoma Lines
RPMI supports NCI-60 cancer lines, Jurkat, Raji, K562, HL-60, and hematopoietic cancer lines where DMEM would alter proliferation and signaling.
Immune Cell OoC
0.04 µm filtered RPMI for tumor-immune interaction chips, vascular-immune OoC, and lymph-node-on-chip models where particle-free media is preferred.
CAR-T & TIL Expansion
Low endotoxin release specification (< 0.05 EU/mL) helps reduce the risk of LPS-driven T cell activation artefacts in CAR-T manufacturing and TIL expansion protocols.
Immune Cell Metabolic Flux
Defined RPMI 1640 base supports extracellular flux analysis, glycolysis stress tests, and ¹³C isotope tracing of lymphocyte activation states. Not compatible with Agilent Seahorse XF assays, which require bicarbonate-free, phenol red-free medium.
Flow Cytometry & Confocal
Ultra-low particulate filtration; phenol red-free variants are available on request to reduce autofluorescence for PE-channel flow cytometry and immune-cell confocal imaging.
