FluxMPS™ RPMI 1640 Medium w/ L-Alanyl-Glutamine, 25mM HEPES: 1X Liquid
FluxMPS™ DCP-RPMIYAH1X is a Microfluidics Suitable, ultra-filtered RPMI 1640 with L-Alanyl-Glutamine + 25mM HEPES formulation engineered for T cells and related immune-cell models on organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. Processed through a quadruple-stage filtration system (0.1 µm ×2 + 0.04 µm ×2), it reaches a 0.04 µm final cut-off — five times finer than the 0.22 µm membranes used for conventional sterile filtration. Formulation: [+] L-Alanyl-L-Glutamine, [+] Sodium Bicarbonate, [+] Phenol Red, [+] 25mM HEPES, [+] Calcium, [+] Magnesium, [+] 2 g/L Glucose, [+] Sodium Pyruvate. HEPES (25 mM, pKa 7.3 at 37°C) supplements the sodium bicarbonate buffering system for added pH stability.
- RPMI 1640 with L-Alanyl-Glutamine + 25mM HEPES — L-Alanyl-L-Glutamine (dipeptide, CAS 39537-23-0) replaces free L-glutamine for stable, non-degrading nitrogen delivery
- Glucose: 2000 mg/L (2.0 g/L); Sodium Pyruvate: 110 mg/L
- 25 mM HEPES (pKa 7.3 at 37°C) supplements the sodium bicarbonate buffering system, reducing pH drift during open-bench handling and flow cytometry sampling
- Quadruple-stage filtration (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final cut-off, with mycoplasma-retentive filtration at every stage
- Endotoxin release specification: < 0.05 EU/mL (LAL assay, USP <85>)
- Manufactured under an ISO 13485:2016 quality management system; final QC at Diagnocine, Totowa NJ

Engineered where standard media fails
Conventional 0.22 µm–filtered RPMI passes mycoplasma, subvisible particulates, and endotoxin fragments that confound sensitive immune-cell culture applications. FluxMPS™ addresses these failure modes with four-stage filtration to a 0.04 µm final cut-off.
Microchannel-safe purity
0.04 µm final filtration with USP <788> Method 1 particulate compliance supports safe perfusion in microfluidic chip geometries.
Specialized dipeptide formulation
L-Alanyl-L-Glutamine (dipeptide, CAS 39537-23-0) replaces free L-glutamine. Use at the same molar concentration as free glutamine (2 mM equivalent). Do not add additional free L-glutamine unless specifically required.
Ultrapure-grade water
Type 1 water, 18.2 MΩ·cm feed water — low trace-metal and low-TOC formulation background for sensitive cell culture outcomes.
Endotoxin below TLR4 threshold
< 0.05 EU/mL endotoxin release specification — below levels associated with TLR4-mediated inflammatory activation in sensitive immune-cell assays.
HEPES pH stability
25 mM HEPES supplements the sodium bicarbonate buffering system, reducing pH drift during open-bench handling and sampling outside the CO2 incubator.
Customization on demand
pH, concentration, component additions, and nutrient modifications available. Contact support@diagnocine.com.
Quadruple-stage filtration system
Four serial filtration stages, run as two dedicated prefilter + final-filter pairs, reaching a final 0.04 µm polish.
-
1
0.1 µm Prefiltration I
Removes large aggregates and cell debris; protects the first 0.04 µm cartridge.
-
2
0.04 µm Final filtration I
Retains sub-micron particulates and mycoplasma-sized organisms (0.2–0.3 µm) that pass a 0.22 µm filter.
-
3
0.1 µm Prefiltration II
Second dedicated prefilter, protecting the second 0.04 µm cartridge.
-
4
0.04 µm Final filtration II — Polish
Ultimate polishing filter; aseptic fill & finish in an ISO Class 5 (Class 100) environment.
Performance vs. conventional media
A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

CAR-T & TIL Manufacturing
Stable L-Alanyl-L-Glutamine avoids ammonia accumulation over multi-week expansion protocols without mid-process replenishment.
Primary Cell Culture
Mycoplasma-retentive filtration and < 0.05 EU/mL endotoxin support sensitive primary immune-cell models.
Metabolic Flux Analysis
Defined formulation base for 13C isotope tracing. Not compatible with Agilent Seahorse XF assays, which require bicarbonate-free, phenol red-free medium.
Microscopy & Optical Sensing
Ultra-low particulate baseline supports confocal microscopy and biosensor OoC applications. Note: this formulation contains phenol red, which may interfere with certain fluorescence/absorbance-based assays.
