Contains L-Glutamine Contains Sodium Bicarbonate Contains HEPES (25 mM) Contains Calcium Contains Magnesium Contains High Glucose Without Phenol Red Without Sodium Pyruvate

FluxMPS™ DCP-RPMIGH-PR1X is a Microfluidics Suitable, Quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) 1X liquid RPMI 1640 medium engineered for organ-on-a-chip (OoC) and microphysiological system (MPS) platforms. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration.

  • Quadruple-stage filtration (0.1 µm ×2 + 0.04 µm ×2) reaching a 0.04 µm final polish, engineered for microfluidic and organ-on-a-chip platforms
  • High-glucose (4500 mg/L) RPMI 1640 base with 25 mM HEPES buffering and 2000 mg/L sodium bicarbonate for dual-buffer pH stability
  • Formulated without sodium pyruvate and without phenol red — suited to fluorescence/absorbance imaging and user-controlled carbon-flux studies

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm–filtered media passes mycoplasma-sized organisms, subvisible particulates, and endotoxin fragments that confound sensitive assays.

Purity Architecture

Quadruple-stage filtration system

Four serial filtration stages — arranged as two dedicated prefilter/final-filter pairs — reach a final 0.04 µm polish under ISO Class 5 aseptic conditions.

  1. 1

    0.1 µm Prefiltration I

Removes large aggregates, cell debris and protein aggregates; provides validated 0.1 µm mycoplasma-retentive filtration (not tested per lot) and protects the first 0.04 µm final filter cartridge.

  2. 2

    0.04 µm Final filtration I

First 0.04 µm pass; retains sub-micron particulates and microaggregates that pass a standard 0.22 µm filter.

  3. 3

    0.1 µm Prefiltration II

Second dedicated 0.1 µm mycoplasma-retentive prefilter, protecting the second 0.04 µm final filter cartridge.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate 0.04 µm polishing filter; ISO Class 5 aseptic fill & finish. Final product QC release gate.

Performance vs. conventional media

5×
Cleaner than 0.22 µm media by particulate count
0.04
µm Final pore size across 4 filtration passes
Sterility & Mycoplasma: No growth after 14-day sterility incubation (USP <71>). Mycoplasma risk is mitigated by 0.1 µm mycoplasma-retentive filtration at every production lot; not independently tested per lot unless stated on the Certificate of Analysis.
Grade: This product is Microfluidics Suitable, filtered to a 0.04 µm final cut-off. It is not an MPS Grade product — that designation is reserved for the 0.01 µm ultra nano-filtered line, which adds 0.02 µm and 0.01 µm stages after the 0.04 µm polish. For applications requiring the 0.01 µm cut-off, contact support@diagnocine.com.

FAQ

Frequently asked questions

Common questions about FluxMPS™ DCP-RPMIGH-PR1X and its use in OoC and MPS applications.

Yes. DCP-RPMIGH-PR1X is specifically engineered for MPS and OoC platforms. The 0.04 µm final filtration helps prevent microfluidic channel fouling, making it suitable for OoC, ToC, BoC, and LoC applications with sensitivity to particulate contamination.
FluxMPS uses a Quadruple-stage system (0.1 µm ×2 + 0.04 µm ×2), providing approximately 5× lower particulate counts and 0.1 µm mycoplasma-retentive filtration that standard 0.22 µm filtration cannot achieve.
This formulation omits phenol red, eliminating background fluorescence and absorbance interference for imaging and biosensor assays, and omits sodium pyruvate, allowing users to control redox and anaplerotic carbon flux. Supplement with 1 mM sodium pyruvate if required for your cell line's cloning efficiency or plating density; add phenol red only if visual pH monitoring is needed for open, non-imaging workflows.
Yes. The sodium bicarbonate (2000 mg/L) buffering system is formulated for 5% CO2 to maintain pH 7.4; the additional 25 mM HEPES provides extra buffering capacity during bench handling outside the incubator.
Yes. This is a basal medium formulation. Supplement with FBS, serum replacements (e.g., B27, N2), or growth factors as required by your cell type and protocol. Pre-filter serum and protein-containing additions through a 0.2 µm low-protein-binding PES or PVDF membrane; do not use a 0.04 µm membrane for serum, as it will strip serum of the proteins and lipoproteins it needs to function.
Endotoxin is controlled per manufacturing batch via LAL assay (USP <85>, Bacterial Endotoxins Test; assay sensitivity 0.005 EU/mL) and must meet the release specification of < 0.05 EU/mL before the batch is released.
Yes. CoA is available per lot upon request at support@diagnocine.com. It includes pH, osmolality, endotoxin, sterility, mycoplasma filtration status, USP <788> particulate data, appearance, and full raw material lot traceability.
