By Henderson–Hasselbalch calculation, maintaining physiological pH requires approximately 8% CO₂ in the incubator atmosphere. The included 15 mM HEPES provides supplemental buffering during bench-top handling outside the incubator but does not eliminate the CO₂ requirement.
