FluxMPS™ Dulbecco's Modified Eagle Medium (DMEM), High Glucose & 25mM HEPES w/o Sodium Pyruvate, Sodium Bicarbonate: 1X Liquid

Contains L-Glutamine Contains Phenol Red Contains 25 mM HEPES Contains Calcium Contains Magnesium Contains High Glucose (4500 mg/L) Without Sodium Bicarbonate Without Sodium Pyruvate

FluxMPS™ DCP-DMEMH-PB1X is a Microfluidics Suitable, quadruple-stage ultra-filtered (0.1 µm ×2 + 0.04 µm ×2) DMEM High Glucose (4.5 g/L) formulation with 25 mM HEPES buffer, engineered for organ-on-a-chip (OoC), microphysiological systems (MPS), and CO₂-independent or open-top microfluidic platforms. A quadruple-stage train (0.1 µm ×2 + 0.04 µm ×2) reaches a 0.04 µm final cut-off, five times finer than the 0.22 µm membranes used for conventional sterile filtration. HEPES (25 mM, pKa 7.3 at 37°C) provides robust CO₂-independent pH buffering. Formulation: [+] High Glucose (4500 mg/L), [+] L-Glutamine, [+] 25mM HEPES, [+] Phenol Red | [-] Sodium Pyruvate, [-] Sodium Bicarbonate.

  • High Glucose (4.5 g/L) — supports neurons, cardiomyocytes, cancer lines, and aerobically active cells
  • 25 mM HEPES (pKa 7.3 at 37°C) — robust, bicarbonate-free pH buffering independent of CO₂ tension
  • Quadruple-stage filtration: Prefiltration I (0.1 µm) → Final filtration I (0.04 µm) → Prefiltration II (0.1 µm) → Final filtration II — Polish (0.04 µm)
  • Contains phenol red (15.9 mg/L) as a pH indicator — orange-colored, clear solution
  • Endotoxin release specification < 0.05 EU/mL (LAL, USP <85>), controlled per manufacturing batch
  • Manufactured under an ISO 13485:2016 quality management system; final QC at Diagnocine, Totowa, NJ
  • Custom pH, salts, glucose, and HEPES concentration adjustments available on request

Why FluxMPS™

Engineered where standard media fails

Conventional 0.22 µm–filtered DMEM passes mycoplasma-scale particulates and subvisible aggregates that clog microfluidic channels. Standard DMEM also lacks HEPES buffering, causing pH instability whenever chips are handled outside incubators. FluxMPS™ addresses both failure modes simultaneously.

filter_alt

Microchannel-safe purity

0.04 µm final filtration; USP <788> particulate compliance supports safe perfusion in sub-100 µm channel geometries.

bolt

High-energy cell support

4.5 g/L glucose sustains energy-demanding cell types — primary neurons, iPSC-derived cardiomyocytes, Warburg-active cancer lines — in long-duration perfusion.

water_drop

HEPES: CO₂-stable pH

25 mM HEPES (pKa 7.3 at 37°C) maintains pH 7.2–7.4 regardless of CO₂ fluctuation — useful for open-top chips, point-of-care devices, and atmospheric incubation.

visibility

Low background for imaging

Quadruple-stage 0.04 µm filtration maintains a low particulate baseline for confocal microscopy and biosensor platforms on chip. This formulation contains phenol red (15.9 mg/L); phenol-red-sensitive fluorescence or absorbance assays should account for background signal.

Purity Architecture

Quadruple-stage filtration system

Four serial filtration stages — two dedicated prefilter + final-filter pairs — reaching a final 0.04 µm polish, delivering sub-mycoplasma-scale purity for microfluidic channels beyond what conventional 0.22 µm media provides.

  1. 1

0.1 µm Prefiltration I — Large Particulate Removal

Removes large aggregates and contaminants; protects the first 0.04 µm cartridge and downstream chip geometries.

  2. 2

    0.04 µm Final filtration I

Retains mycoplasma-scale particulates (0.2–0.3 µm) and sub-micron aggregates that pass a standard 0.22 µm filter.

  3. 3

    0.1 µm Prefiltration II

Second dedicated prefilter, protecting the second 0.04 µm cartridge.

  4. 4

0.04 µm Final filtration II — Polish

Ultimate polishing filter; ISO Class 5 aseptic fill & finish.

FAQ

Frequently asked questions

Common questions about FluxMPS™ DCP-DMEMH-PB1X DMEM High Glucose + 25mM HEPES.

Yes. DCP-DMEMH-PB1X combines quadruple-stage 0.04 µm filtration for ultra-low particulate delivery in microfluidic channels with 25 mM HEPES buffering for CO₂-stable pH. High glucose (4.5 g/L) supports energy-demanding cell types — neurons, cardiomyocytes, and cancer lines — in long-duration chip perfusion. The HEPES component is particularly valuable for open-top chips where CO₂ concentration fluctuates.

Sodium bicarbonate is removed so HEPES alone controls pH without CO₂; sodium pyruvate is removed for fresh researcher-defined supplementation. This provides a CO₂-independent, high-glucose base with L-Glutamine already included — ideal for atmospheric, open-top microfluidic platforms where pyruvate concentration and timing are experimental variables.
No. This formulation is CO₂-independent — HEPES (25 mM) alone maintains pH 7.2–7.4 at 37°C without gas supplementation. HEPES (pKa 7.3 at 37°C) maintains stable pH across a broad CO₂ range, making this medium suitable for open-top chips, point-of-care platforms, and atmospheric incubation where CO₂ control is impractical.
Yes. Add FBS (typically 5–10%), serum-free supplements, growth factors, antibiotics, or custom nutrients as required. Filter serum-containing additions through a 0.2 µm low-protein-binding PES or PVDF membrane; do not use a 0.04 µm membrane for serum, as it retains lipoproteins and much of the functional serum fraction. Contact support@diagnocine.com for custom co-formulation.
Endotoxin is controlled per manufacturing batch by LAL assay (USP <85>, assay sensitivity 0.005 EU/mL). Every batch is tested before release and must meet the specification: < 0.05 EU/mL.
Yes. A full CoA per batch covers: appearance, pH (USP <791>), osmolality (USP <785>), sterility (USP <71>), endotoxin (USP <85>), mycoplasma filtration control, particulate count (USP <788> Method 1), and raw-material traceability. Request at support@diagnocine.com.
