Description
CD247, also known as CD3Z (T-cell surface glycoprotein CD3 zeta chain), belongs to the CD3Z/FCER1G family. CD3 is a complex of proteins that directly associates with the T cell receptor (TCR). CD3 is considered to be a pan-T cell marker for detection of normal and neoplastic T cells. Probable role in assembly and expression of the TCR complex as well as signal transduction upon antigen triggering.

Human Diseases
- Immunodeficiency 25: Associated with defects in T-cell function.
- T-B+ Severe Combined Immunodeficiency: Results from mutations affecting CD3 subunits including CD3 zeta.
Cellular Signaling Pathways
- Plays a critical role in T-cell activation and signal transduction through the T-cell receptor (TCR).
- Phosphorylation of ITAMs (Immunoreceptor Tyrosine Activation Motifs) leads to ZAP70 activation.
- Involved in synapse formation between T cells and antigen-presenting cells.
