Description:
The 49.5% Acrylamide:Bisacrylamide Solution (32:1) is a specialized, high-concentration reagent used in molecular biology and biochemistry for protein and nucleic acid electrophoresis. This solution contains 48 g of acrylamide and 1.5 g of N,N'-methylenebisacrylamide (bis-acrylamide) per 100 mL, resulting in a total concentration (T) of 49.5% and a crosslinker concentration (C) of 3%. The 32:1 ratio of acrylamide to bis-acrylamide offers a unique balance, producing gels with slightly larger pore sizes compared to the more common 29:1 ratio. This formulation is particularly suitable for separating larger proteins or nucleic acid fragments, potentially providing enhanced resolution for certain molecular weight ranges. As a ready-to-use solution, it eliminates the need for handling hazardous powdered acrylamide, improving laboratory safety and ensuring consistent gel preparation. Researchers using this solution should optimize their electrophoresis protocols to account for its specific properties, as gel characteristics may differ from those made with standard ratios.
Advantages:
- Larger pore sizes: Compared to the more common 29:1 ratio, the 32:1 ratio produces gels with slightly larger pore sizes.
- Enhanced resolution for larger molecules: The larger pore sizes make this solution particularly suitable for separating larger proteins or nucleic acid fragments.
- Flexibility in gel preparation: The 32:1 ratio allows researchers to optimize their electrophoresis protocols for specific applications, especially when working with larger biomolecules.
- Ready-to-use formulation: As a pre-mixed solution, it eliminates the need to handle hazardous powdered acrylamide, improving laboratory safety and ensuring consistent gel preparation.

All DiagnoCine Precision Sterile buffers are filtered-sterilized with a 0.1-micron filtration two times and 0.04-micron filtration one time. Thus, mycoplasma contamination is prevented. The smallest size mycoplasma type can be about 0.2 microns.
